You can't get to PJ via Midtown Direct, alas. There are two options:

1) Take a Midtown Direct train to Newark, then take a local back to Hoboken, and finally, take a train from Hoboken to PJ.
2) Take the PATH from Midtown to Hoboken and hop on a PJ train.

Matamoras is right across the Delaware from Port Jervis. There are taxis at PJ, according to Switchboard:
